efficiency of systemic versus intralesional bone marrow-derived stem cells in regeneration of oral mucosa after induction of formocresol induced ulcers in dogs

چکیده
background: bone marrow mesenchymal stem cells (bmscs) are the key to regenerative wound healing. mscs have spatial memory and respond to local environment. the goal of this study was to evaluate the use of systemic and intralesional transplantation of bmscs for regeneration of oral mucosa in an in vivo dog model. materials and methods: transplantation of undifferentiated green fluorescent protein (gfp)- labeled autologous bmscs systemically, submucosally or vehicle (saline) was injected around the chemically induced oral ulcer in each group of 18 adult dogs. the healing process of the ulcer was monitored clinically and histopathologically. gene expression of vascular endothelial growth factor (vegf) and collagen genes was detected in biopsies from all ulcers. one way anova was used to compare between means of the three groups. results were considered significant at p < 0.05. results: flow cytometric analysis of the mscs at the passage 3 showed that these cells were negative for cd45 (2.39%). they expressed high levels of cd29 (98.34%). frozen fluorescence microscopy of sections of the cell-treated oral tissue of all groups indicated that the gfp-transduced implanted cells were integrated within the transplanted tissues. the treatment resulted in dramatic wound edge activation and resurfacing of oral mucosa wound. conclusion: our results revealed that bmscs may be labeled with (gfp), in order to know the distribution of these cells after administration, and suggest that intralesional administration is an appropriate procedure to achieve acceptable regeneration of the previously injured oral mucosa more than systemic route. key words: bmscs, intralesional, oral ulcer, systemic

The present study determined the regenerative effect of bone marrow-derived stem cells (BMSCs) on thioacetamide (TA)-induced liver fibrosis in rats. A total of 30 male Wistar rats were randomly divided into sham control and treatment groups. The rats of the sham control group were subdivided into three groups and sampled on the 14th, 18th, and 20th weeks after fibrosis induction. The rats of th...

BACKGROUND Infertility is a serious social problem in advanced nations, with male factor in half of all cases of infertility. This study was conducted to determine the regenerative effect of bone marrow-derived stem cells in spermatogenesis of infertile hamster. METHODS Twelve adult male hamsters were equally divided into azoospermic and control groups. Busulfan was intraperitoneally used fo...
متن کاملEmergence of signs of neural cells after exposure of bone marrow-derived mesenchymal stem cells to fetal brain extract
Objective(s): Nowadays much effort is being invested in order to diagnose the mechanisms involved in neural differentiation. By clarifying this, making desired neural cells in vitro and applying them into diverse neurological disorders suffered from neural cell malfunctions could be a feasible choice. Thus, the present study assessed the capability of fetal brain extract (FBE) to induce rat bon...

Objective(s):Bone marrow-derived mesenchymal stem cells (BMSCs) have attracted significant interest to treat asthma and its complication. In this study, the effects of BMSCs on lung pathology and inflammation in an ovalbumin-induced asthma model in mouse were examined. Materials and Methods:BALB/c mice were divided into three groups: control group (animals were not sensitized), asthma group (an...
